When crocheting from word chart and I turn my work, do I skip the first stitch?

Welcome!
Your turning chain doesn't count as an actual stitch, so no, don't skip the first stitch. While some conventional (non-graph) patterns sometimes call for counting the turning chain, some don't, and in this case it's inadvisable to count it, as your stitching will no longer work with the graph. You also shouldn't work into the turning chain when you get to the end of the row, as this will also throw your work off of the graph. Just keep single crochets to single crochets and you should be good to go!

So when I get to the end of a row I should chain one but not count it as part of the actual pattern?

count as a stitch; only your actual stitches count.
